I'm having trouble understanding the step with the overlay/screen layers, though... Is that something that can only be done in paintoolSAI? (I'm using Photoshop...) Do you color what you want shiny on the overlay layer only? Is there anything in the screen layer?

Nope! It can be done in Photoshop as well!
Hrm, how can explain this.
Try testing it out yourself so you'll know what I mean:
(Try opening a finished work first)
We have two layers right? The first one is the overlay layer and the one on top should always be the screen layer. Note that these two should always be on top of all the layers you have.
Coloring on the overlay layer using an airbrush, depending on what color you choose, changes the hue/shade of the color below, depending on how much opacity you put on the brush. (You can sometimes get a darker or lighter hue)
The purpose of the screen layer is to make that area glow
Screen+Overlay=glowing magic
I like using oranges, reds, apple green, really bright colors since they are quite an eye candy
